Metabolic Coupling Mechanisms Between Stromal Cells and Immune Cells in Tertiary Lymphoid Structures

Tertiary lymphoid structures (TLS) are ectopic lymphoid tissues formed under pathological conditions such as tumors, chronic inflammation, and autoimmune diseases. Their maturation status is closely associated with the intensity of antitumor immune responses. TLS are composed of B cell zones, T cell zones, follicular dendritic cells, fibroblastic reticular cells, and high endothelial venules. Their formation and function are profoundly influenced by the metabolic microenvironment. This review systematically summarizes the metabolic coupling mechanisms between stromal cells and immune cells within TLS, focusing on three levels: key nutrient exchange, metabolite mediated signal transduction, and the construction of spatial metabolic niches. These mechanisms drive TLS maturation and provide new intervention targets for enhancing antitumor immunity.
